چکیده
To whet your appetite for the special focus in next issue’s Leading Edge on Genomic Instability, we take a look at what’s new in the field of DNA repair. Ranging from network analyses of the proteins involved in mutagenic DNA repair to examination of single molecules taking part in homologous recombination, this Select probes the inner workings of a process that can wreak havoc during carcinogenesis while remaining vital to the generation of a functioning T cell repertoire. Tune in again on January 31 for Reviews on: aneuploidy, recombination in the immune system, telomere biology, and what ciliates can teach us about programmed genome instability.

It is now universally accepted that DNA is the main target for damages caused by physical and chemical genotoxicants. Although there are different methods to measure directly the induced DNA damages but due to fast repair processes in cellular environment, most of the damages would be repaired even before sampling, therefore processed DNA damages, i.e. damages left unrepaired after acting repai...
متن کاملI-38: Chromosome Instability in The Cleavage Stage Embryo
Recently, we demonstrated chromosome instability (CIN) in human cleavage stage embryogenesis following in vitro fertilization (IVF). CIN not necessarily undermines normal human development (i.e. when remaining normal diploid blastomeres develop the embryo proper), however it can spark a spectrum of conditions, including loss of conception, genetic disease and genetic variation development. To s...
متن کاملI-37: Genome Instability and DNA Damage in Male Somatic and Germ Cells Expressed as Chromosomal Microdeletion and Aneuploidy Is A Major Cause of Male Infertility
Background: Sperm chromatin insufficiencies leading to low sperm count and quality, infertility and transmission of chromosomal microdeletion and aneuploidies to next generations can be due to exposure to environmental pollutions, chemicals and natural or manmade ionizing radiation. In this project which has continued for more than 10 years and is unique in many technical aspects in Iran and in...
متن کاملPCNA Retention on DNA into G2/M Phase Causes Genome Instability in Cells Lacking Elg1
Loss of the genome maintenance factor Elg1 causes serious genome instability that leads to cancer, but the underlying mechanism is unknown. Elg1 forms the major subunit of a replication factor C-like complex, Elg1-RLC, which unloads the ring-shaped polymerase clamp PCNA from DNA during replication. Here, we show that prolonged retention of PCNA on DNA into G2/M phase is the major cause of genom...
متن کاملLeveraging DNA Damage Response Signaling to Identify Yeast Genes Controlling Genome Stability
Oncogenesis frequently is accompanied by rampant genome instability, which fuels genetic heterogeneity and resistance to targeted cancer therapy. We have developed an approach that allows precise, quantitative measurement of genome instability in high-throughput format in the Saccharomyces cerevisiae model system. Our approach takes advantage of the strongly DNA damage-inducible gene RNR3, in c...
متن کاملTelomere-related Genome Instability in Cancer
tumor types are in an astonishing state of disarray. The extent of genome scrambling was only appreciated after development of high-resolution techniques. Spectral karyotyping (M-FISH, SKY) has painted a picture of extensive reshuffling of chromosome segments. Techniques that display the differences between normal and cancer genomes, combined with DNA microarrays (array-CGH, Pinkel et al. 1998;...
